Components in this kit are prepared with pure chemicals according to our proprietary technology. BioChain’s Mitochondria Activity Assay (Cytochrome C Oxidase Activity Assay) Kits are designed to measure the cytochrome c oxidase activity in mitochondria sample, as well as the integrity of mitochondria. Different amounts of cytochrome c oxidase positive control were assayed to define the linear range of enzyme activity for the assay: 0.35 milliunit to 3.5 milliunit. One kit is consisted of reagents enough for performing 100 assays.

Description


Mitochondrial activity in an isolated subcellular fraction can be measured by assaying for mitochondrial specific enzymes. The mitochondria activity assay kit is designed to measure the mitochondria-specific cytochrome C oxidase activity in soluble and membrane bound mitochondria samples. Cytochrome C has a sharp absorption band at 550nm in the reduced state.  Upon oxidation, this band becomes weaker and broader. This colourimetric assay is based on observation of the decrease in absorbance at 550nm of ferrocytochrome C caused by its oxidation to ferricytochrome C by cytochrome C oxidase. It is typical to determine the extent of reduction of cytochrome C by measuring the difference in absorbances at 550nm and 565nm. For optimal preparation of mitochondria, we recommend using

The Mitochondria Isolation Kit meets the demand for an effective method to isolate intact mitochondria (and mitochondrial protein) from tissues and cultured cells. The kit allows for the isolation of intact and active mitochondria in a fast, simple and cost effective process.  MitoLight® Mitochondrial Apoptosis Detection Kit

 

The MitoLight™ Apoptosis Detection Kit utilizes a lipophilic cation, termed as MitoLight™, as a mitochondrial activity marker. MitoLight™ is a mitochondrial dye that stains mitochondria in living cells in a membrane potential-dependent fashion. The monomer is in equilibrium with so-called J-aggregates, which are favored at higher dye concentration or higher mitochondrial membrane potential.
MitoLight™ partitions differently in healthy cells than in apoptotic cells. Therefore, it has been possible to use a fluorescence ratio-ing technique to study mitochondrial membrane potentials. In healthy cells, the dye accumulates and aggregates in the mitochondria, giving off a bright red fluorescence (l em = 585-590 nm). In apoptotic cells with altered mitochondrial membrane potential, the dye in its monomeric form stays in the cytoplasm, fluorescing green (l em = 527-530 nm), providing a ready discrimination between apoptotic and nonapoptotic cells. The fluorescence can be observed by fluorescence microscopy using a band-pass filter (detects FITC and rhodamine) or analyzed by flow cytometry using FITC channel for green monomers (Ex/Em = 488/530) and PI channel for red aggregates (Ex/Em = 488/585).
